NEFA C Assay Kit
Wako announces the availability of its NEFA C test
kit for the quantitative determination of non-esterified (or free)
fatty acids in serum or plasma (non-heparinized). The Wako NEFA
C assay utilizes an in vitro enzymatic colorimetric methodology,
which eliminates the need for an extraction procedure and can be
automated using your existing clinical chemistry instrumentation.
The Wako enzymatic method relies upon the acylation of coenzyme
A(CoA) by the fatty acids in the presence of added acyl-CoA synthetase
(ACS). The acyl-CoA produced is oxidized by added acyl-CoA oxidase
(ACOD) with the generation of hydrogen peroxide. Hydrogen peroxide,
in the presence of peroxidase (POD) permits the oxidative condensation
of 3-methyl-N-ethyl-N-(b-hydroxyethyl)-aniline (MEHA) with 4-aminoantipyrine
to form a purple color which can be measured spectrophotometrically
at 550nm. The Wako NEFA C assay has excellent performance. The within-run
precision is 2.7%CV, 1.1%CV and 1.1%CV for mean concentrations of
0.33mmol/L, 0.62mmol/L and 0.99mmol/L, respectively. The linear
range of the assay is 0-2.0 mmol/L
